Distinct myeloid cell populations induce allogeneic mixed leukocyte reactions of differing intensity. Three cell populations (CD14−CD16++, CD14+CD16+, CD14+CD16low/-) were purified as shown in Figure 2C, irradiated and cultured at the indicated cell ratios with responder PBMC (105 per well) for 5 days. Proliferation was measured by the incorporation of methyl-3H thymidine ([3H]TdR; 0.5 μCi per well) for the final 18 h of culture. Data are presented as the corrected counts per minute (ccpm) averaged over 3 min. Error bars denote ± SE of the technical replicates and letters represent statistical significance between proliferation induced by each cell population for each responder:stimulator ratio for p < 0.05. One representative experiment of three is shown.
