Interactions between different HN proteins and vimentin. HD11 cells in 6-well cell culture plates were transfected with HN-expressing (pcDNA3.1-JHN or pcDNA3.1-MHN) or empty (pcDNA3.1) plasmid. Transfected cells were harvested and lysed 24 hpt, and the experiment was conducted according to the manufacturer’s instructions for the Co-Immunoprecipitation Kit (Santa Cruz Biotechnology, USA). The immunoprecipitated proteins were then identified and analysed by Western blotting using an anti-HN antibody, an anti-vimentin antibody or normal mouse/rabbit IgG. Three groups of cells were included in the co-IP experiment: the input group, antibody group and IgG group. pcDNA3.1-JHN: pcDNA3.1 plasmid containing the sequence encoding the JS/7/05/Ch-type HN protein. pcDNA3.1-MHN: pcDNA3.1 plasmid containing the sequence encoding the Mukteswar-type HN protein.
