Various dose of TAK-242 inhibited the rTsgal-activated MAPK/NF-κB pathway. Caco-2 cells were first treated with TLR-4 inhibitor TAK-242 (1–25 µM) for 2 h, and then incubated with rTsgal, IIL CAg and LPS for 0.5 h, and Western blot was performed to analyze the expression level of p-ERK1/2, p-NF-κB p65 and NF-κB p65. A Western blotting of NF-κB pathway proteins of Caco-2 cells treated with TAK-242. B Expression of p-ERK1/2 in TAK-242 treated-Caco-2 cells. Expression of rTsgal-activated p-NF-κB p65 (C) and NF-κB p65 (D) in Caco-2 cells was inhibited by various dose of TAK-242. *P < 0.0001 compared to the PBS group.
